Unusually massive line of storms aim at Midwest

WASHINGTON (AP) ? A gigantic line of powerful thunderstorms could affect one in five Americans on Wednesday as it rumbles from Iowa to Maryland packing hail, lightning and tree-toppling winds.

Meteorologist are warning that the continuous line of storms may even spawn an unusual weather event called a derecho (duh-RAY'-choh), which is a massive storm of strong straight-line winds spanning at least 240 miles. Wednesday's storms are also likely to generate tornadoes and cause power outages that will be followed by oppressive heat, said Bill Bunting, operations chief at the National Weather Service's Storm Prediction Center in Norman, Okla.

The risk of severe weather in Chicago, Indianapolis, Cincinnati and Columbus, Ohio, is roughly 45 times higher than on a normal June day, Bunting said. Detroit, Baltimore, Washington, Milwaukee, Pittsburgh and Louisville, Ky., have a risk level 15 times more than normal. All told, the area the weather service considers to be under heightened risk of dangerous weather includes 64 million people in 10 states.

"It's a pretty high threat," Bunting said, who also warned that the storms will produce large hail and dangerous lightning. "We don't want to scare people, but we want them to be aware."

Wednesday "might be the worst severe weather outbreak for this part of the country for the year," said Jeff Masters, meteorology director at Weather Underground.

You can have tornadoes and a derecho at the same time, but at any given place Wednesday the straight-line winds are probably more likely.

Last year, a derecho caused at least $1 billion in damage from Chicago to Washington, killing 13 people and leaving more than 4 million people without power, according to the weather service. Winds reached nearly 100 mph in some places and in addition to the 13 people who died from downed trees, another 34 people died from the heat wave that followed in areas without power.

Derechoes, with winds of at least 58 mph, occur about once a year in the Midwest. Rarer than tornadoes but with weaker winds, derechoes produce damage over a much wider area.

Wednesday's storm probably won't be as powerful as 2012's historic one, but it is expected to cause widespread problems, Bunting said.

The storms are the type that will move so fast that "by the time you see the dark sky and distant thunder you may have only minutes to get to safe shelter," Bunting said.

The storms will start late morning or early afternoon in eastern Iowa, hit Chicago by early afternoon and move east at about 40 mph, Bunting said. If the storm remains intact after crossing the Appalachian Mountains, which would be rare for a derecho, it should hit the Washington area by late afternoon or early evening, he said.

For Washington, Philadelphia and parts of the Mid-Atlantic the big storm risk continues and even increases a bit Thursday, according to the weather service.

Well, it pays to stick around a launch venue as the cleanup crews begin to roll out. We just caught a glimpse of Sony's brand new PlayStation 4 following the company's press event tonight, and it's really quite stunning. We were only able to view the console from a right angle, but we did manage to make out a dark black glossy finish up top, with a gray matte panel just below. It's difficult to judge depth from the side, but from this front view at least, we definitely liked what we saw.

A small silver PlayStation logo is positioned towards the bottom of the glossy panel, representing the only branding visible from this side of the device. There's a Sony logo on the left side as well, as you can see in our gallery below. It's really quite a sleek design, so even though you may opt to toss the console away in a cabinet, it'll look just fine sitting out, too. Unfortunately Sony staffers ushered us out after snapping this shot, so that's all we've got for now. We'll be hitting up the company's booth bright and early tomorrow morning, though, and you better believe we'll be back to share our full hands-on impressions then.

Despite?or maybe because of?their perfectionist approach to egalitarianism, lesbian couples seem to be more likely to break up than gay ones. Pepper Schwartz noted this in the early 1980s, as did the 2006 study of same-sex couples in Sweden and Norway, in which researchers speculated that women may have a ?stronger general sensitivity to the quality of relationships.? Meaning maybe women are just picky, and when you have two women, you have double the pickiness. So perhaps the real threat to marriage is: women.

Epigenetic factor likely plays a key role in fueling most common childhood cancer

June 10, 2013 ? Changes in an epigenetic mechanism that turns expression of genes on and off may be as important as genetic alterations in causing pediatric acute lymphoblastic leukemia (ALL), according to a study led by scientists at St. Jude Children's Research Hospital and published in the June 10 online edition of the Journal of Clinical Investigation.

The results suggest the mechanism called cytosine methylation plays a previously under-appreciated role in the development of leukemia. Cytosine methylation involves adding or removing methyl groups to cytosine, which is a building block of DNA.

The study is the most comprehensive effort yet to identify and understand genetic and epigenetic factors that work together to cause ALL, the most common childhood cancer. ALL is a cancer of white blood cells known as lymphocytes. Scientists at St. Jude and Weill Cornell Medical College collaborated on the project.

Researchers used a variety of techniques to examine hundreds of thousands of methylation sites across the genome in normal and leukemic lymphocytes, including samples from more than 160 children with ALL. Investigators found that known ALL subgroups, which are defined by chromosomal alterations, have unique methylation profiles. Those profiles correlated with different patterns of gene expression.

"It is well known that different leukemia subgroups have distinct patterns of gene expression that are important in the development of leukemia," said Charles Mullighan, MBBS (Hons), MSc, M.D., an associate member of the St. Jude Department of Pathology. Mullighan and Ari Melnick, M.D., Gebroe Professor Hematology/Oncology at Weill Cornell Medical College, are the study's co-corresponding authors.

"We have assumed that the underlying genetic changes are important determinants of those gene expression profiles. We now know that changes in methylation state also have key roles in influencing gene expression," Mullighan said.

The study used tissue samples from 137 St. Jude patients with B-cell leukemia and 30 children with T-cell leukemia. The patients represented all major ALL subgroups.

"The data show that aberrant epigenetic gene programming can now be considered a hallmark of acute lymphoblastic leukemia, occurring in all patients regardless of the presence of genetic mutations," Melnick said. "This offers the opportunity for development of epigenetic targeted therapies for patients with ALL that could be broadly applicable to many patients."

For comparison, researchers also checked B and T cells from 27 healthy children. Investigators found that leukemia cells shared a core group of abnormally methylated genes. The genes included ones involved in regulating the cell division and proliferation. "This remains to be tested, but the findings suggest that alterations in methylation are an important early step in the development of leukemia," Mullighan said.

The research provides further evidence that genetic and epigenetic events are both important in establishing different subgroups of ALL. For this study, researchers conducted genome-wide sampling of methylation, gene expression and DNA structural abnormalities, including the gain or loss of DNA. Shann-Ching Chen, Ph.D., St. Jude Pathology, developed many of the methods used to integrate and analyze the results. Chen and Maria Figueroa, now of the University of Michigan and formerly of Cornell, are the study's co-first authors.

The study also found that more than one-third of 71 genes targeted by genetic alterations are also abnormally methylated in ALL. The methylation changes involved known tumor suppressor or oncogenes genes including CDKN2A, CDKN2B, PTEN and KRAS. "The findings suggest these genes are inactivated or deregulated more frequently than suggested by simply analyzing structural changes in the genome," Mullighan said.

The other authors are Anna Andersson, formerly of St. Jude and now of Lund University Hospital, Sweden; Letha Phillips, formerly of St. Jude; Mondira Kundu and James Downing, both of St. Jude; Yushan Li, Cornell; and Jason Sotzen, University of Michigan.
